John's father was Thomas Barlow and his mother was Betty (Elizabeth) Mather. His maternal grandparents were Thomas Mather and Ann Entwistle. He had four brothers and three sisters, named William, Thomas, Robert, James, Maria, Jane and Ann. He was the youngest of the eight children. He died as an infant in 1844 in Holcombe.²
