Isabel was born in Pembroke, Pembrokeshire, Wales.  Isabel's father was Richard FitzGilbert ("Strongbow") de Clare and her mother was Aoife MacMurrough of Leinster.  Her paternal grandparents were Earl Gilbert FitzGilbert ("Strongbow") de Clare of Pembroke and Isabel de Beaumont; her maternal grandparents were King Dermot MacMurrough and More O'Toole.  She was an only child.  She died in 1220 in Pembroke, Pembrokeshire, Wales.  She was buried in Tintern Abbey, Tintern, Monmouthshire, England.

‌Isabel and William were married in a religious ceremony in August 1189 in London, Middlesex, England.  They had five daughters named Maud, Eve, Joan, Isabella and Sibyl.
