Samuel's father was Thomas Hopley and his mother was Rebecca Jones. His paternal grandparents were Thomas Hopley and Jane Shone. He had two brothers and three sisters, named Thomas, Randle, Elizabeth, Margaret and Mary. He was the fifth oldest of the six children.
