יעקב was born in 1902.  יעקב's father was אברהם ישראל מרדכי גליס and his mother was ברכה באשע אסתר סופר.  His paternal grandparents were יעקב המנקר גליס and אסתר מינדל פרס; his maternal grandparents were שמעון אליהו צבי סופר and חיה נחמה בת אברהם.  He had a brother and four sisters, named ניסן צבי הירש, רבקה מייטא, פייגע ליבא, ציפורה and שרה רחל - שרהל'ה.  He was the third oldest of the six children.
