יעקב המנקר was born in 1850 in גרודנו, פולין.  יעקב המנקר's father was ישראל גליס and his mother was רבקה לא ידוע.  His paternal grandparents were צבי גליס and רבקה מייטא בת רב קלמן מהורודנא.  He had a brother named דוד לייב.  He died at the age of 63 on May 29th, 1913 in ירושלים.
