Karel was born in 1738.  Karel's father was Jan Nepomuk Mitrovský z Nemyšle and his mother was Marie Kazimíra Blankovská z Dorušic.  His paternal grandparents were Arnošt Matyáš Mitrovský z Nemyšle and Marie Terezie Lehocká. He had two brothers named Josef Antonín and Antonín Benjamin.  He was the youngest of the three children.  He died at the age of 78 in 1816.
