Jan Nepomuk's father was Arnošt Matyáš Mitrovský z Nemyšle and his mother was Marie Terezie Lehocká.  His paternal grandparents are Karel Mitrovský z Nemyšle and Helena Sobek-Kornitz. He had four brothers and four sisters, named Maxmilian Josef, Jan Baptista, Gottfried, Arnošt Benjamin, Anna Eleonora, Barbora, Johana Josefa and Helena Konstancie.  He died in 1760.
