Euphrosyne was born in Constantinople, Byzantine Empire.  Euphrosyne's father was Theodorus Kastamonites and her mother was Margaretha Von Ungarn.  Her maternal grandparents were Bela III Von Ungarn and Agnes De Chatillon-sur-Loing. She had two half-brothers named Manuel and John.  She died after 1195 in Constantinople.

‌Euphrosyne and Andronikos Dukas were married in a religious ceremony in Constantinople, Turkey.  They had six sons and three daughters, named Theodoros, Ionnis, Andronikos, Konstantinos, Alexios III Komnenos, Isaakios II, Irini, Zoe and Theodora.
