Agnes De was born about 1250 in Of, Derby, Derbyshire, England.  Agnes De's father was William de Ferrers 7 E Derby and her mother was Margaret De Quincy.  Her paternal grandparents were William de Ferrers Earl of Derby and Agnes De Meschines Lady of Chartley; her maternal grandparents were Roger Quincy Earl of Wincester and Helen (Elena) Galloway. She had two brothers and two sisters, named Robert, William de, Joan De and Elizabeth.  She is the youngest of the five children. She had a half-brother and six half-sisters, named John De, Maud (Matilda) De, Isabel de, Joan De, Sibyl De, Agatha De and Eleanor De.
