Isabel de Clare was born in of Pembroke, Wales.1 Isabel de Clare's father was Richard FitzGilbert de Clare Earl of Pembroke and her mother was Aoife (Eva) MacDermot Cts de Ireland. Her paternal grandparents were Gilbert FitzGilbert de Clare Earl of Pembroke and Isabel (Elizabeth) de Beaumont; her maternal grandparents were Diarmait MacMurrough King of Leinster and Mor ingen Muirchertaig O'Toole.
