Karl was born in 1730. Karl's father was August Anhalt-Köthen, von and his mother was Emilie Promnitz, von. His paternal grandparents were Emanuel Lebrecht Anhalt-Köthen, von and Gisela Agnes Rath, von. He had two brothers and two sisters, named Friedrich, Friedrich Erdmann, Christiane Anna and Wilhelmine.
