Wilhelm Ernst was born in 1717.  Wilhelm Ernst's father was Ernst August I. Sachsen-Weimar-Eisenach, von and his mother was Eleonore Wilhelmine Anhalt-Köthen, von.  His paternal grandparents were Johann Ernst III. Sachsen-Weimar, von and Sofie Auguste Anhalt-Zerbst, von; his maternal grandparents were Emanuel Lebrecht Anhalt-Köthen, von and Gisela Agnes Rath, von. He had two brothers and five sisters, named Johann Wilhelm, Emanuel Friedrich, Wilhelmine Auguste, Charlotte Agnes, Johanna Eleonore, Ernestine Albertine and Bernhardine Christiane.  He was the oldest of the eight children. He had four half-brothers and a half-sister, named Karl August, Ernst August II., Ernst Adolf, Ernst Friedrich and Ernestine Sophie Auguste.  He died at the age of 2 in 1719.
