Charlotte was born in 1744. Charlotte's father was Friedrich Karl Holstein-Plön, von and her mother was Christiane Irmgard Reventlow, von. Her paternal grandparents were Christian Karl Holstein-Norburg, von and Dorothea Christine Aichelberg, von. She had a brother and three sisters, named Christian Karl, Sofie Christine Luise, Friederike and Luise. She was the fourth oldest of the five children. She died at the age of 26 in 1770.
