Ann was born about 1530 in Of Salmesbury, Lancashire, England. Ann's father was Thomas Southworth and her mother was Margery Boteler. Her maternal grandparents were Thomas Boteler and Margaret Delves. She had a brother and five sisters, named John, Margaret (Ludworth), Elizabeth, Cicely, Katherine and Dorothy.
