Isabel was born about 1254 in Thorpe Arnold, Leicestershire, England. Isabel's father was Arnold IV Du Bois. Her paternal grandparents are Ernald III Du Bois and Joan De Beauchamp. She had two brothers named John and William. She was the second oldest of the three children. She died about 1282.
