Samuel was born on January 1st, 1747 in , , Coleraine, Ireland. Samuel's father was John KNOX and his mother was Jeane GRACY. His paternal grandparents are Ad KNOX and Margaret Brock; his maternal grandparents are John GRACEY and Jean Sinclair. He had six brothers and a sister, named William, Absalom, John, Joseph, James, Benjamin and Mary. He was the fourth youngest of the eight children.
