Desire was born in 1625 in Plymouth, Mass.. Desire's father was John Howland and her mother was Elizabeth Tilley. Her paternal grandparents were Henry Howland and Margaret Anne Aires; her maternal grandparents were John Tilley and Joan Rogers Hurst.
