Duncan was born in 1564 in Lochaber & Ardchoille,Scotland and his baptism took place there in 1564. Duncan's father was Gregor MacGregor and his mother was Isabel Cameron. His paternal grandparents were Duncan Ladafach MacGregor and Mary Campbell; his maternal grandfather was Unknown Cameron. He was an only child.
