Isabel de's father was Juan Diego Sánchez de Morales and her mother was Inés de Leon Carvajal.  Her paternal grandparents were Juan Morales de Negrete and Inés de Rodriguez. She had two half-sisters named Isabel de and Luisa.

They had a daughter named Maria.
