Rupilla's father was Rupilius Libo Frugi and her mother was Salonia Matidia.  Her maternal grandparents were Salonia Matidius and Ulpia Marciara Traiana.  She was an only child.

They had a son and two daughters, named M. Annius, Annia Galeria and Annia Vera.
