Rodrigo Manrique's father was Alonso Manrique and his mother was Leonor Acuña.  His paternal grandparents were Rodrigo Manrique Conde De Paredes and Elvira De Castañeda.  He was an only child.

They had a daughter named Sebastiana Manrique.
