Dorothea was born in 1623. Dorothea's father was August Anhalt-Köthen, von and her mother was Sibylle Solms-Laubach, von. Her paternal grandparents were Joachim Ernst Anhalt, von and Eleonore Württemberg, von. She had three brothers and four sisters, named Ernst Gottlieb, Lebrecht, Emanuel, Johanna, Ehrenpreis, Sofie and Elisabeth. She was the fourth oldest of the eight children. She died at the age of 14 in 1637.
