Duraisamy's father is Savariappan and his mother is Chinnammal.  His paternal grandfather was Anthonimuthu Goundar and his paternal grandmother is <Unknown>; his maternal grandparents are Rajendiran (Kanniyappan), Narthampattiyan and Maria Soosaiyamma. He had four brothers and two sisters, named Jeganathan, Masilamani, Arputham, Pushpanathan, Anthoniammal and Therese Mary.  He is the youngest of the seven children. He has two half-brothers and two half-sisters, named Arockiam Malaysia, Periyakai Selvanayagam, Kottampatti Aaya Ambrose and Kitheru.

Duraisamy and Amala are married.  They have two sons and four daughters, named Arulraj, Gnananantham, Diraviyam, Poongavanam, Geetha and <Unknown>.
